张幸鼎

个人简介:

现任中山大学医学院教授,博士生导师。于2004年开始攻读硕士和博士学位期间主要在苏州大学特聘教授秦正红教授研究组进行神经退行性疾病中自噬通路的研究工作。2011年博士毕业后进入了苏州大学唐仲英医学研究院血液学研究中心杨林教授实验组进行博士后研究,2012年2月赴美国MD安德森肿瘤中心Dr.Orlowski教授实验室接受博士后训练。2017年初获得美国著名大学西北大学研究助理教授职位并成立实验室。依托于芝加哥安&罗伯特·卢瑞儿童医院的平台,建立了以小儿脑瘤模型为研究中心,并包含了其他小儿肿瘤研究的科研平台。

已在神经科学和肿瘤研究领域的国际知名杂志上发表SCI论文22篇,其中第一作者和通讯作者11篇,包括Cancer Cell (IF: 23.324),Autophagy (IF: 9.108), Cell death & disease (IF: 5.378),总引用率大于 700次,独立他引次数超过400次。

2017年6月获聘中山大学“百人计划”引进人才。

重要学术研究成果与贡献:

(1)在国际上第一次提出TJP1蛋白作为骨髓瘤化疗的标志物,并对TJP1蛋白介导化疗耐药性的机制做出了深入探讨。相关结果以第一作者研究论文发表在国际肿瘤知名杂志Cancer Cell (IF: 23.324)上。著名科学类网站Science Daily, Medicalxpress和美国临床肿瘤学会ASCO都进行了详细报道。哈佛大学医学院的资深骨髓瘤专家Dr.Mitsiades教授在同期的Cancer Cell杂志上对本文进行了深入探讨并称赞了本文的重要的基础和临床意义。更重要的是,此部分的研究成果对于骨髓瘤病人的临床化疗靶点检测以及对化疗的指导作用已经在世界第一的肿瘤中心安德森肿瘤中心进入临床实际的检测和应用。

(2)完善了大鼠纹状体急性线粒体损伤的模型的建立,首次提出了p53和相关蛋白在急性线粒体损伤和兴奋性损伤中的作用。利用这些新的实验方法和实验设想,一系列自噬相关通路在线粒体和兴奋性损伤模型和脑缺血模型中被首次发现。此部分的结果于2008和2009年发表在Autophagy (IF:9.108) 杂志3篇高水平研究论文,迄今为止得到了接近400次的引用。

(3)创新性的发现自噬和凋亡蛋白DRAM1,HMGB1和Bcl-2在调节大鼠纹状体急性线粒体损伤的作用。之前研究发现p53调节的DRAM1,HMGB1和Bcl-2基因在肿瘤中发挥了调节作用,但没有发现此通路对于神经性推行疾病的作用。申请人创新性的在线粒体损伤的模型中对信号通路进行研究,发现DRAM1,HMGB1和Bcl-2蛋白调节了自噬和凋亡途径来保护线粒体损伤导致的神经退行性疾病的发展,填补了相关研究的空白。此部分的结果以第一作者和通讯作者发表在Cell Death Dis (IF: 5.378), PLoS One (IF:3.057) 和J Neurosci Res(IF:2.689) 杂志上并受到了134次世界范围的广泛引用。
